Dear Participants of MAD-MS Study,

Your participation in the MAD-MS study has led to groundbreaking discoveries that are reshaping our understanding of multiple sclerosis. Together, we’ve revealed how chronic MS lesions expand due to persistent inflammation, discovered the critical role of the choroid plexus as a new biomarker for disease progression, and provided the first human evidence linking chronic demyelination to nerve fiber loss. These findings, published in 36 papers across leading medical journals including Brain and Lancet Neurology, are already influencing clinical trials and treatment approaches worldwide. The measurement tools we developed are now being used globally to test new therapies and monitor disease progression more effectively.

Building on the insights gained from MAD-MS, we have designed a clinical trial called COMPASS-MS. This retrospective study aims to evaluate the impact of current disease-modifying therapies (DMTs) on smouldering inflammation and disease progression in MS. By leveraging existing data from multiple sites, COMPASS-MS will generate actionable insights within 12–18 months, significantly faster than traditional prospective trials.

The study will provide a comprehensive assessment of which therapies are most effective in addressing progressive aspects of MS. These findings have the potential to transform lives by identifying treatments that can slow or prevent the accumulation of disability caused by chronic inflammation.

While we are currently working to secure funding for COMPASS-MS, our team remains fully committed to advancing this critical research.
